introduction | 2-chloro-3-fluoro-isonicotinic acid is an organic intermediate, which can be prepared from 2-chloro-3-fluoropyridine by reaction with strong alkali and dry ice. |
use | 2-chloro -3-fluoro-isonicotinic acid is a carboxylic acid organic substance, and it has been reported in the literature that it can be used to prepare ADP receptor antagonist antiplatelet drugs. |
preparation | n-butyl lithium (290ml,0.72mol) is added to diisopropylamine (95ml,0.66mol) in a solution of 500ml anhydrous THF under nitrogen at -78 ℃. After stirring for 0.5 hours, the resulting mixture was added to a solution of 2-chloro-3-fluoropyridine (78g,0.60mmol) in 200ml of anhydrous THF at -78°C. after stirring for 3 hours, pour the mixture into dry ice (340g,6.0mol). Dilute the mixture with water (1L) and adjust the pH to about 2 with 2M aqueous hydrochloric acid. The product was extracted with ethyl acetate (3 × 500mL), and the combined extract was washed with brine, dried with Na2SO4, filtered and concentrated, and the solid residue was recrystallized from EtOAc to give the title compound. |
